Digitonin perfusion of rat liver
نویسنده
چکیده
Perfusing a rat liver with digitonin in the concentration range 2-20mg/ml results in complete decolorization of the organ within 45-250s. Decolorization progresses with time in the direction of flow, and it is therefore possible, by collecting the eluate, to obtain material from specific intracellular compartments of hepatocytes in different zones in the microcirculatory unit of the liver. The results demonstrate that cytoplasmic marker enzymes from periportal or perivenous hepatocytes can be collected with as little contamination from the other compartment as is obtained in microdissection studies. Furthermore, a fraction enriched in mitochondrial marker enzymes can be achieved with only 10-20% contamination by cytoplasmic material.

Digitonin perfusion in the study of metabolic zonation of the rat liver: potassium as an intracellular concentration reference.
The mammalian liver microcirculation seems to be uniquely organized in order to create maximum difference of the biochemical milieu to which the upstream (periportal) and downstream (perivenous) parenchymal cells are exposed. Indeed the existence of a subdivision of hepatocytes in zones of different metabolic function, long predicted by enzyme zonation studies (for a review, see Jungermann & Ka...
متن کاملTitration of rat liver with digitonin: a well defined short term damage of cellular metabolism.
Carefully performed pulse titration of the isolated rat live in the course of continuous erythrocyte free perfusion with small amounts of digitonin causes a short term permeability of liver cell plasma membranes with concomitant short lived release of intracellular low or high molecular compounds such as ATP or lactate dehydrogenase. Gluconeogenesis from lactate being completely inhibited durin...

Intact rat liver cells from the perivenous region were isolated by collagenase perfusion after first destroying the periportal region by a brief portal infusion of digitonin. Periportal cells were isolated after retrograde digitonin infusion. Significantly higher alanine aminotransferase, gamma-glutamyltransferase and lactate dehydrogenase activities and lower glutamate dehydrogenase and pyruva...
متن کاملDual - digitonin - pulse perfusion
A previously described digitonin-perfusion technique [Quistorff, Grunnet & Cornell (1985) Biochem. J. 226, 289-297], by which intracellular material of rat liver could be liberated, has been refined, now allowing release of cytosol of high purity from both periportal and perivenous parts of the same liver. The cytosolic fractions are obtained by perfusing the liver for short intervals (10-20 s)...

liver injury induced by viruses, chemicals and drugs can be protected by different medicinal plants. Feijoa sellowiana (Myrtaceae) is an evergreen bush native to southern areas of South America, as well as Iran where the fruits are very popular. Feijoa has shown a potent antimicrobial effect. Morever, the antioxidant activity of total Feijoa extract has also been reported. MDMA or ecstasy is a ...
